چکیده

Two-dimensional thin-layer chromatography was used to resolve cyclophosphamide metabolites present (a) in blood and urine of cyclophosphamide-treated rats and (b) after incuba tion of cyclophosphamide with hepatic microsomes, a hepatic 9000 X g supernatant fraction, or a model oxygenase system. Incubation of cyclophosphamide with rat hepatic microsomes yielded a single metabolite (aldophosphamide) capable of alkylating 4-(/>-nitrobenzyl)pyridine. This metabolite could be trapped as the semicarbazone by the addition of semicarbazide to the incubation mixture. Aldophosphamide was unstable to incubation at 37°for 44 hr, degrading to

Bioassay and relative cytotoxic potency of cyclophosphamide metabolites generated in vitro and in vivo.

Cytotoxic potencies of cyclophosphamide, 4-ketocyclophosphamide, carboxyphosphamide, mechlorethamine (HN2), bis(2-chloroethyl)amine (nor-HN2), chlorambucil, acrolein, and cyclophosphamide metabolites generated in vivo and in vitro were determined via bioassay. Our bioassay procedure was to incubate the potential cytotoxic agent with Walker 256 ascites cells in vitro, inject these cells into hos...

The Protective Effects of Origanum vulgare L. Extract on Genetic Damage of Cyclophosphamide in Mice Blood Lymphocytes Using Micronucleus Test

Despite its various clinical applications, cyclophosphamide (CP), an alkylating chemotherapeutic agent, has demonstrated numerous side effects, including genetic toxicity. This study investigated the protective action of Origanum vulgare L., a powerful antioxidant plant, on the genotoxicity of CP in the mice blood lymphocytes. The mice were pretreated orally with different doses of (50, 100, 20...

The Effect of Vitamin E and Selenium on Cyclophosphamide Detoxification in Hepatic Tissues of Mature Rats

Introduction and Objective: liver is an important body organ without which life is impossible. Cyclophosphamide is an alkylating agent widely used as an anti-cancer (“antineoplastic” or “cytotoxic”) chemotherapy drug. The present study aimed to find a suitable antioxidant and examine the effect of vitamin E and selenium on reducing the side effects of cyclophosphamide on liver as a vit...

De novo expression of transfected human class 1 aldehyde dehydrogenase (ALDH) causes resistance to oxazaphosphorine anti-cancer alkylating agents in hamster V79 cell lines. Elevated class 1 ALDH activity is closely correlated with reduction in DNA interstrand cross-linking and lethality.

Human class 1 aldehyde dehydrogenase (hALDH-1) can oxidize aldophosphamide, a key aldehyde intermediate in the activation pathway of cyclophosphamide and other oxazaphosphorine (OAP) anti-cancer alkylating agents. Overexpression of class 1 ALDH (ALDH-1) has been observed in cells selected for survival in the presence of OAPs. We used transfection to induce de novo expression of human ALDH-1 in ...
